    The relationship between web enjoyment and student perceptions and learning using a web-based tutorial

    Web enjoyment has been regarded as a component of system experience. However, there has been little targeted research considering the role of web enjoyment alone in student learning using web-based systems. To address this gap, this study aims to examine the influence of web enjoyment on learning performance and perceptions by controlling system experience as a variable in the study. 74 students participated in the study, using a web-based tutorial covering subject matter in the area of 'Computation and algorithms'. Their learning performance was assessed with a pre-test and a post-test and their learning perceptions were evaluated with a questionnaire. The results indicated that there are positive relationships between the levels of web enjoyment and perceived usefulness and non-linear navigation for users with similar, significant levels of system experience. The implications of these findings in relation to web-based learning are explored and ways in which the needs of students who report different levels of web enjoyment might be met are discussed

    Emotional Reaction of Experiencing Crisis in Virtual Reality (VR)/360° Immersive technology is being used to provoke emotion and move millions of people to action. Because organizations and filmmakers are exploring more ways to use the immersive technology of VR and 360-degree video to evoke emotion, it is important to investigate what emotional reactions are experienced. Both VR and 360° fall under the category of immersive media and the terms are used interchangeably in this study. NVivo 11 is used for the analysis of 1,700 Twitter texts between the years 2015 and 2017 after people view the crisis Clouds Over Sidra in VR/360°. The appraisal theory of emotion serves as the framework to explore the interpretation of the subject’s emotional reactions. Sentiment and thematic analysis reveal (a) an increase in empathy, (b) reports of emotional reactions including feelings of sadness, grief and anger, (c) greater understanding of the crisis (d) intentions to act related to the crisis, (e) importance of VR/360° for educational use, and (f) the power of VR/360° and its ability to alter fields of education, humanitarian work, and politics. This study finds the immersive experience of viewing a crisis in VR/360° generates a range of highly emotional reactions. It is an important goal to understand the role VR/360° plays in generating emotional reactions and behavioral change, particularly in view of the accelerating development of emotional VR/360° content and people’s access to immersive technology

    BACKGROUNDThe benefits of using a head tracker in a fast jet to aim weapons and control sensor direction have been amply demonstrated, but head direction is difficult to control at high G. An experiment was conducted to assess whether aiming using the eye, rather than the head, might be advantageous.METHODSA centrifuge provided sustained accelerations of up to 8 Gz. Participating aircrew were asked to point at static targets using head- or eye-aiming, maintain the acquisition while the target was lit, and then transfer to the next target as quickly as possible. A helmet-mounted scene camera recorded the subject's view of the target board. A laser mounted by the scene camera was used for head-aiming and to determine head direction. The eye tracker camera and illuminator were also helmet-mounted. The eye and scene images were recorded and analyzed off-line.RESULTSEye-aiming acquisition times and accuracy were affected only slightly by Gz; however, both head-aiming speed and accuracy deteriorated as Gz increased. Eye-aiming was substantially faster than head-aiming at all Gz levels, but head-aiming was more accurate under these experimental conditions. Subjective ratings supported the objective data. A majority of subjects preferred eye to head as an aiming mechanism.CONCLUSIONSEye-aiming was faster and easier to use than head-aiming at all Gz levels, and particularly at high Gz

    A protective Zika virus E-dimer-based subunit vaccine engineered to abrogate antibody-dependent enhancement of dengue infection

    Infections with dengue (DENV) and Zika (ZIKV) viruses can induce cross-reactive antibody responses. Two immunodominant epitopes, to precursor membrane protein (prM) or the fusion loop epitope (FLE) on envelope (E) protein are recognized by cross-reactive antibodies1, 2, 3 that are not only poorly neutralizing, but can also promote increased viral replication and disease seerity via Fc-gamma receptor mediated infection of myeloid cells, a process termed antibody-dependent enhancement (ADE)1, 4, 5 . ADE is a significant concern for both ZIKV and DENV vaccines as the induction of poorly-neutralizing cross-reactive antibodies may prime an individual for ADE upon natural infection. In this report, we describe the design and production of covalently-stabilized ZIKV E-dimers, which lack prM and do not expose the immunodominant FLE. Immunization of mice with ZIKV E-dimers induces dimer-specific antibodies, which protected against ZIKV challenge during pregnancy. Importantly, the ZIKV E-dimer-induced response does not cross-react with DENV or induce ADE of DENV infection

    Understanding genetic control of root system architecture in soybean : insights into the genetic basis of lateral root number

    Developing crops with better root systems is a promising strategy to ensure productivity in both optimum and stress environments. Root system architectural traits in 397 soybean accessions were characterized and a high-density single nucleotide polymorphisms (SNPs)-based genome-wide association study was performed to identify the underlying genes associated with root structure. SNPs associated with root architectural traits specific to landraces and elite germplasm pools were detected. Four loci were detected in landraces for lateral root number (LRN) and distribution of root thickness in diameter Class I with a major locus on chromosome 16. This major loci was detected in the coding region of unknown protein, and subsequent analyses demonstrated that root traits are affected with mutated haplotypes of the gene. In elite germplasm pool, 3 significant SNPs in alanine-glyoxalate aminotransferase, Leucine-Rich Repeat receptor/No apical meristem, and unknown functional genes were found to govern multiple traits including root surface area and volume. However, no major loci were detected for LRN in elite germplasm. Nucleotide diversity analysis found evidence of selective sweeps around the landraces LRN gene. Soybean accessions with minor and mutated allelic variants of LRN gene were found to perform better in both water-limited and optimal field conditions

    Human monoclonal antibodies against chikungunya virus target multiple distinct epitopes in the E1 and E2 glycoproteins

    Chikungunya virus (CHIKV) is a mosquito-transmitted alphavirus that causes persistent arthritis in a subset of human patients. We report the isolation and functional characterization of monoclonal antibodies (mAbs) from two patients infected with CHIKV in the Dominican Republic. Single B cell sorting yielded a panel of 46 human mAbs of diverse germline lineages that targeted epitopes within the E1 or E2 glycoproteins. MAbs that recognized either E1 or E2 proteins exhibited neutralizing activity. Viral escape mutations localized the binding epitopes for two E1 mAbs to sites within domain I or the linker between domains I and III; and for two E2 mAbs between the β-connector region and the B-domain. Two of the E2-specific mAbs conferred protection in vivo in a stringent lethal challenge mouse model of CHIKV infection, whereas the E1 mAbs did not. These results provide insight into human antibody response to CHIKV and identify candidate mAbs for therapeutic intervention
